Primary Applications
1. Plant Genetic Transformation Research: As a specific chromogenic substrate for the Gus reporter gene, it forms a deep blue, insoluble precipitate visible to the naked eye when catalyzed by β-glucosidase. This allows for direct observation of the expression location and intensity of exogenous genes in transgenic plants, making it the most classic staining and identification reagent in plant molecular biology experiments.
2. Microbial Detection: Used for color-based screening of bacterial colonies—such as E. coli—that contain the uidA gene. When added to microbial culture media, positive colonies turn deep blue, significantly improving the efficiency of screening for positive strains.
3. Histochemical experiments: Used for in situ enzyme activity staining of animal and plant tissue sections to precisely localize the expression distribution of β-glucosidase within tissue cells; widely applied in research experiments related to developmental biology and pathological testing.
Standard Storage Requirements
1. Safety and Isolation Requirements: Must be stored strictly separate from strong oxidizing agents and strong acids. Keep away from heat sources and direct sunlight to prevent oxidation and decomposition of the product, which would result in the loss of its color-forming activity.
2. Operational Control Guidelines: Wear a lab coat, disposable gloves, and safety goggles during handling. Avoid direct inhalation of dust or prolonged skin contact. Dispose of waste reagents in accordance with regulations for biochemical laboratory waste; direct discharge into the natural environment is strictly prohibited.
