Purpose:

L-Glycinolide hydrochloride, as an important intermediate, is widely used in organic synthesis and is also a key intermediate in the synthesis of the chiral herbicide L-glufosinate.

Preparation:

It can be prepared from the natural amino acid L-methionine. The commonly used method is a three-step process: first, L-methionine reacts with iodomethane to form a hydride salt; then, under basic conditions, this is converted to L-homoserine; and finally, under acidic conditions, dehydration occurs to yield the L-homoserine lactone hydrochloride. Since this method involves numerous separation steps, some studies have attempted a “one-pot method” to simplify the process, in which intermediate products are not separated, and the final product is purified only in the last step.
